Total cell lysates (15 µg total protein) from PC-3, HepG2, SH-SY5Y (reduced expression control), and HaCat (reduced expression control) cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-FOXP1 Antibody, clone O94D2, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker. Predicted expression data was obtained from Human Protein Atlas.

Description

FOXP1 (Forkhead Box P1) is a transcription regulator, which plays an important role in the brain, cardiac, lung, and lymphocyte development. Mutations in FOXP1, which are located at 3p13, have been reported in patients with global developmental delay (GDD), intellectual disability (ID), and speech defects. FOXP1 is targeted by recurrent chromosome translocations and its overexpression confers a poor prognosis in a number of types of lymphomas, suggesting it may function as an oncogene. In contrast, FOXP1 localizes to a tumor suppressor locus at 3p14.1 and the loss of FOXP1 expression in breast cancer is associated with a worse outcome, suggesting FOXP1 may function as a tumor suppressor in other tissue types. Recent studies also suggest that in embryonic stem cells (ESC), specific isoforms of FOXP1 help maintain the ESC pluripotency and stimulates the expression of transcription factor genes that are required for pluripotency such as OCT4, NANOG, NR5A2, and GDF3, while concomitantly repressing genes required for ESC differentiation.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone is not recommended for ChIP (Chromatin Immunoprecipitation) assays (as determined by in-house testing).
