Total cell lysates (15 µg total protein) from PC-3, A549 and HaCat cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-Artemis Antibody, clones 15E7C90 and 13A9C90, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker. Cell lysates were loaded in order of increasing DCLRE1C mRNA expression levels (source: Human Protein Atlas).

Description

Artemis, also known as DCLRE1C, is a nuclear protein belonging to the metallo-β-lactamase superfamily. Artemis has been found to be required for the assembling of B cell antibody genes and T cell receptor genes from individual V, D, and J segments. During V(D)J recombination, Artemis forms a complex with DNA-dependent protein kinase (DNA-‘PK) and cuts the DNA with hairpin structure. Deficiency in Artemis results in blocking of V(D)J recombination and failure to produce mature B and T cells, which is known as the condition of severe combined immune deficiency (SCID). Artemis is also found to be involved in the termini processing step of double-strand break (DSB) repair. Artemis-deficient cells are more sensitive to DSB induced by ionizing radiation or chemical agents such as bleomycin and neocarzinostatin.
