SK-BR-3 cells were fixed with 4% Fixation Buffer (Cat. No. 420801) for 10 minutes and permeabilized with 0.5% Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with 5.0 µg/mL of purified anti-CD340 (ErbB2/HER-2) (clone A20145B), followed by incubation with Alexa Fluor® 594 goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801) (panel B). Images were captured with a 60X objective. Scale bar: 50 µm

Description

CD340 is the protein HER2, a member of the epidermal growth factor receptor (EGFR) family of cell membrane tyrosine kinases, and is encoded by the gene ErbB2. Unlike other EGFR family members, CD340 contains no ligand binding domain and has no known ligands. Instead, this protein interacts with other EGF receptor family members to form a heterodimer, stabilizing ligand binding and enhancing kinase-mediated downstream signaling. CD340 has been shown to be involved in embryonic development and cancer progression; this protein is amplified in adenocarcinoma of the lung, some breast cancers, glioblastoma, gastric cancer, and ovarian carcinoma, where it functions as an oncogene. Overexpression of ErbB2 in breast cancers has been shown to confer Taxol resistance and is a therapeutic target in a number of human cancers.

Recommended Usage

Each lot of this antibody is quality control tested by immunocytochemistry. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This product is not recommended for use in IHC-P.

Antigen Details

Structure
CD340 is a 1,255 amino acid protein with a predicted molecular weight of 137.9 kD.
Distribution

Expressed on the cell surface of many tumor cells including some breast cancers, lung adenocarcinoma, gastric cancer, ovarian cancer, glioblastoma, some c-ALL blasts and bone marrow MSCs

Function
Contains no ligand binding domain and interacts with other EGF receptor family members to form a heterodimer, stabilize ligand binding, and enhance kinase-mediated downstream signaling. Involved in embryonic development and cancer progression.
Interaction
CD340 interacts with a large number of proteins, including SHC1, EGF receptor, neuregulin 1, c-Src, integrin β4, Grb2, Grb7, SOS1, caveolin-1, JAK2, PAK1, FAK, and β-catenin, among others.
Ligand/Receptor
Despite being a member of the epidermal growth factor receptor (EGFR) family, no ligand of CD340 has been identified.
