    HUVEC cells (positive cell line, panel A) and HeLa cells (negative cell line, panel B) were fixed with Fixation Buffer (Cat. No. 420801) for 10 minutes followed by permeabilization with 100% ice-cold methanol for 10 minutes, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with 10 µg/mL of Purified anti-CD102 (ICAM-2) (clone W22039C), followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 60X objective. Scale bar: 50 µm

    Whole cell extracts (15 µg total protein) from indicated cell lines were incubated at 37°C for 30 minutes and were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.5 µg/mL of Purified anti-CD102 (ICAM-2) (clone W22039C) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607903) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

Description

CD102 (ICAM-2) is a 55 kD type I transmembrane glycoprotein which belongs to the immunoglobulin superfamily. It is also known as Intercellular Adhesion Molecule-2, or ICAM-2. CD102 is constitutively expressed on vascular endothelial cells, some lymphocytes, monocytes, and platelets. CD102 is thought to be involved in lymphocyte recirculation, and in co-stimulation of the immune response. CD102 and CD54 in endothelial cells, such as in blood-brain barrier, are essential for assisting T cell polarization and crawling and diapedesis. CD102 is a counter receptor of LFA-1 (CD11a/CD18), DC-SIGN and MAC-1.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. For immunocytochemistry, a concentration range of 1 - 10 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For western blotting (WB) it is recommended to heat the cell lysate at 37 °C for 30 min rather than boiling it.

For immunocytochemistry (ICC), either Fixation Buffer (Cat. No. 420801) or 4% paraformaldehyde followed by methanol permeabilization is recommended.

The reactivity of this antibody (clone W22039C) in cell surface flow cytometry is able to be blocked by another BioLegend antibody (clone CBR-IC2/2) (Cat. No. 328502).
