    HeLa cells untreated (panels A and B) or treated with 100 ng/mL of PMA (panels C and D) were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with Triton X-100 for 10 minutes, and blocked with 5% FBS for 1 hour. Cells were then intracellularly stained with 5.0 µg/mL of Alexa Fluor® 647 anti-FosB (clone W19011B) (red) overnight at 4°C. Nuclei were counterstained with DAPI (Cat. No. 422801). Panels A and C represent Alexa Fluor® 647 channel; panels B and D represent merged Alexa Fluor® 647 and DAPI channels. Images were captured with a 60X objective. Scale bar: 50 µm

Description

FosB is a member of the FOS family of proteins that dimerizes with proteins of the Jun family to form the AP-1 transcription factor complex. As part of AP-1, FosB plays a critical role in regulating cell differentiation, proliferation, and apoptosis. FosB is expressed in most tissues and has been particularly well-studied in the CNS. ΔFosB, a truncated splice variant of the FOSB gene, plays a critical role in the brain’s reward circuit plasticity, with significant impact on both natural and pathological behavior. FosB also functions as a tumor suppressor, and dysregulation of FosB activity or levels has been found in numerous cancers, including gastric, prostate and colorectal cancer.

Recommended Usage

Each lot of the test-sized antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining using the test sizes, the suggested use of this reagent is 5 µL per million cells in 100 µL staining volume or 5 µL per 100 µL of whole blood. It is recommended that the reagent be titrated for optimal performance for each application.

Each lot of the µg-sized antibody is quality control tested by immunocytochemistry. For immunocytochemistry using the ug size, a concentration range of 1.25 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application. 

* Alexa Fluor® 647 has a maximum emission of 668 nm when it is excited at 633 nm / 635 nm.

Distribution

FosB is widely expressed in developing and adult tissues, including the brain, thyroid, bladder and adipose tissue.

FosB heterodimerizes with proteins of the Jun family to form the transcription factor AP-1 to regulate cell proliferation, survival, and differentiation.
