Description

Glycogen synthase kinase 3 beta or GSK3β, is a 420 aa length constitutively active serine-threonine kinase that phosphorylates more than 100 target proteins. GSK3β is involved in several biological activities including glycogen metabolism, cell signaling, cellular transport, cell proliferation, apoptosis, and others. The activity of GSK3β is regulated by phosphorylation at different residues. Phosphorylation at tyrosine 216 enhances GSK3β kinase activity, while phosphorylation at serine 9 decreases kinase activity. In addition, GSK3β activity can also be regulated by cellular localization and/or formation of protein complexes. GSK3β has been associated with several diseases including type 2 diabetes, Alzheimer’s disease, inflammation, cancer, addiction, and bipolar disorders.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Antigen Details

Structure
GSK3β is a 420 a.a protein with a predicted molecular weight approximately 47 kD. GSK3β has a bi-lobar architecture, with N-terminal and C- terminal domains. The N-terminal is responsible for ATP binding, while the C-terminal carries the activation loop that mediates the kinase activity.
Distribution

GSK3β can be found mainly in the cytoplasm, however, it could also be present in mitochondria and nucleus.  

Function
It is a serine/threonine Kinase, its activity can be enhanced by phosphorylation on its tyrosine 216, or reduced by phosphorylation on its serine 9.
