Whole cell extracts (15 µg total protein) from human brain membrane tissue was heated at 37°C for 30 minutes, resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 μg/mL of purified Ankyrin-G antibody (clone N106/20) overnight at 4 °C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50,000 dilution (lower). Lane M: Molecular weight marker.

Description

Ankryin G is a submembranous scaffold protein expressed in a variety of tissues, including brain, heart, and muscle. In the neurons, Ankyrin G is localized in the axon initial segment and the nodes of Ranvier. Ankyrin G binds to βIV spectrin and actin to create a submembranous cytoskeletal network in axons. By itself, Ankyrin G controls the ion channel clustering and the maintenance of neuronal polarity.
