IHC staining of purified anti-PAR-2 antibody (clone SAM11) on formalin-fixed paraffin-embedded human heart tissue. Following antigen retrieval using Sodium Citrate H.I.E.R (Cat. No. 928602), the tissue was incubated with 10 µg/ml of the primary antibody overnight at 4°C. BioLegend´s Ultra-Streptavidin (USA) HRP kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm

Enhanced chemiluminescence was used as the detection system.

Description

Proteinase-activated receptor 2 (PAR-2), also known as G-protein coupled receptor 1 or thrombin receptor-like 1, is a receptor for trypsin and trypsin-like enzymes coupled to G proteins. The function of PAR-2 is mediated through the activation of multiple signaling pathways, including MAPK, phospholipase C, intracellular calcium, and I-kappaB kinase/NF-kappaB and Rho. PAR-2 is involved in the regulation of innate and adaptive immunity, the modulation of inflammatory responses, and as a sensor of proteolytic enzymes generated during the course of infection. PAR-2 is also involved in the recruitment of leukocytes to inflammation sites, promotion of dendritic cell maturation and trafficking to lymph nodes, production of superoxide, and T-cell activation.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration of 10 µg/ml is suggested. For Western blotting, the suggested use of this reagent is 0.5 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody is effective in formalin-fixed paraffin-embedded immunohistochemical staining (IHC-P) and immunoblotting (WB).
