Total cell lysates (15 µg total protein) from HepG2 and A549 cells (negative control), HEL and NIH/3T3 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-NFATC4 Antibody, clone W17026A, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:10000 dilution (lower). Lane M: Molecular Weight marker. Predicted NFATC4 expression data was obtained from Human Protein Atlas.

Description

NFATC4 is a transcriptional activator that induces expression of cytokine genes, including IL2 and IL4 in activated T cells. It also plays a central role in cardiovascular, muscle, and nervous cell development, differentiation, and function. Under basal conditions, NFATC4 is extensively phosphorylated and retained in the cytosol. Upon stimulation, elevated calcium results in NFATC4 dephosphorylation by calcineurin and subsequent translocation into the nucleus.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.25 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone displayed a weak affinity for mouse NFATC4 when tested by WB.
