Total cell lysates (15 µg total protein) from undifferentiated THP-1 cells (-) and THP-1 cells treated with PMA for 3 days followed by an overnight LPS treatment (+) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 2.0 µg/mL (1:250 dilution) of Purified anti-DDX58 (RIG-1) Antibody, clone 9F1B20, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-Actin Antibody (Cat. No. 664804) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular Weight marker.

Description

DDX58, also named RIG-1, is a member of the DExD/H-box-containing RNA helicase family. DDX58 was first reported as a retinoic acid-inducible gene and subsequently characterized as a pattern recognition receptor (PRR). DDX58 is responsible for sensing cytosolic viral RNA and has been shown to initiate innate antiviral immune responses. DDX58 is composed of two caspase recruitment domains (CARDs), a helicase domain, and a C-terminal regulatory domain (CTD). In response to virus infections, the DDX58 CTD domain binds 5'-triphosphorylated dsRNA, which induces multimerization of DDX58 with the adaptor molecule MAVS on the mitochondria through the CARD domains. MAVS transduces the signal to activate downstream transcription factors, such as IRF-3, IRF-7, and NF-κB, leading to production of proinflammatory cytokines and type I interferons.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.5 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Function
