Whole cell extracts (30 µg protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to PVDF and probed with 1.0 µg/mL (1:500 dilution) purified anti-CTCF antibody (clone W19193A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin antibody (Cat. No. 643807) was used as a loading control at a 1:10000 dilution (lower). Lane M: Molecular weight marker.

Description

CTCF is an 83 kD transcription factor found in the nucleoplasm, which translocates to the nucleolus when the cell is undergoing cell differentiation. This protein can act as an insulator protein by binding to an unmethylated imprinting control region (ICR) structure and prevent the activation of cis-acting elements which in turn prevents gene activation. CTCF becomes further involved in gene regulation when it forms a complex with cohesin. This complex can stabilize the formation of a chromatin loop, which can promote or repress the binding of other transcription factors to the promoter and enhancer, leading to gene activation or gene repression respectively. CTCF has a role as a tumor suppressor, when the gene goes through a mutation or deletion there is a rise in variance in CpG methylation throughout the genome. This methylation contributes to the development of aggressive and invasive tumors which are seen in breast, endometrial and bladder cancers.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 1.0 - 2.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone was tested for ICC on HeLa cells fixed with 4% PFA and permeabilized with either methanol or Triton X-100. Both methods resulted in high cytosolic background staining.
