Total lysates (15µg protein) from 293T and 293T/CDKN2A knockdown (KD) cells were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with 1:500 diluted (1µg/mL) purified anti-CDKN2A (p16) antibody, clone 15C10C30 (upper) or 1:3000 diluted anti-GAPDH (poly6314) antibody (lower). Proteins were visualized by chemiluminescence detection using a 1:3000 diluted goat anti-mouse-IgG secondary antibody conjugated to HRP for the anti-CDKN2A (p16) antibody, and a donkey anti-rabbit IgG Antibody conjugated to HRP for GAPDH. Lane M: Molecular weight ladder, M* indicates longer exposure.

Description

Cyclin-dependent kinase inhibitor 2A (CDKN2A) isoform 1, also known as p16-INK4a, is an inhibitor of the cyclin dependent kinases CDK4 and CDK6. CDKN2A acts as a negative regulator of cell cycle progression by preventing phosphorylation of the retinoblastoma protein (pRb) by cyclin-dependent kinases. CDKN2A expression is upregulated during cellular senescence and aging. It functions as a tumor suppressor to induce cell cycle arrest and aging of normal tissues. Germ line point mutations, homozygous deletion, or promoter methylation in the human CDKN2A locus causes uncontrolled cell proliferation, which results in a variety of malignancies such as melanoma, pancreatic adenocarcinoma, gastric cancer, and esophageal cancer.
