Total lysates (15 µg protein) from Jurkat, Daudi (low expression, negative control) and Raw264.7 were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1:500 (1 ug/ml) purified anti-CD247 antibody, clone 6B10.2. Proteins were visualized using chemiluminescence detection by incubation with and HRP goat anti-mouse IgG secondary antibody (Cat. No. 405306, 1:3000 dilution). Direct-Blot™ HRP anti-β-actin antibody was used as a loading control (Cat. No. 643807, 1:8000 dilution).

Description

The invariant TCR zeta chain is a member of the CD3 complex associated with the clonotypic alpha/beta TCR heterodimer. The disulfide-linked TCR zeta homodimers transmit signals following TCR ligation. Loss of TCR zeta expression has been reported in a diverse range of disease states, including autoimmune diseases, many neoplastic conditions and chronic infections, such as tuberculosis and leprosy.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. Western blotting, suggested working dilution(s): Use 0.5-1 µg per ml antibody dilution buffer. It is recommended that the reagent be titrated for optimal performance for each application.

Structure
Structure: 163 amino acid long protein (predicted MW is 18kDa), with a short extracellular domain of only nine amino acids conserved between mouse and human. There are 6-7 intracellular tyrosines that are potential phosphorylation sites.

T cells and NKT cells. Reported also positive on CD16+ human NK cells.
