IHC staining of Alexa Fluor® 647 anti-MECP2 antibody (clone N227/21) on formalin-fixed paraffin-embedded mouse brain tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with 5 µg/ml of the primary antibody overnight at 4°C. Nuclei were counterstained with Hoechst, and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 10X objective. Scale Bar: 100 µm

Description

Methyl CpG binding protein (MECP2), also known as autism susceptibility to X-linked 2 (AUTSX3), is a nuclear protein crucial for normal neuronal function that binds to methylated DNA, forming complexes with histone deacteylases and turning off gene expression.  MECP2 expression is an X-linked trait. Loss of MECP2 expression causes Rett Syndrome (also known as RTS, RTT, or RS) a neurological disorder which causes decelerated growth of hands, feet, and the head.
