Successfully detects CD4+ T cells in thymus, spleen, and lymph node with high signal.

Experimental Design

Application:	Flow Cytometry
Cells used:	Thymus, spleen, lymph node cells
Brief Protocol:	Extract thymus, spleen, and lymph node from euthanized mouse using tweezers and place in FACS buffer (PBS + 2% FBS + 1mM EDTA) Mash up with frosted slides and treat with Ack lysis buffer to get rid of erythrocytes. Stain cells with Fc block at 1:200 and place on ice for 5 min. Stain cells with anti-mouse CD4-647 antibody at 1:200 to 1:500 and place on ice for 20-30 min. Wash 3x with FACS buffer and complete FACS.
Results Summary:	Successfully detects CD4+ T cells in the thymus, lymph node and spleen.
