CCL5 (RANTES), also known as CCL5, is a 7.8 kD member of the CC family of chemokines, which is expressed in T cells and monocytes/macrophages. RANTES is strongly chemotactic for monocytes/macrophages, memory T cells, eosinophils, and induces histamine release from basophils. In a solution, RANTES can form high molecular weight aggregates that exhibit dissimilar biological activities to its dimeric form. The aggregates cause T cell and neutrophil activation while promoting HIV infection, whereas the dimeric form exhibits RANTES-induced chemotaxis and inhibits HIV infection. As T cells become activated, RANTES secretion appears to decrease.Product Details
- Human RANTES, amino acids (Ser24-Ser91) (Accession# NM_002985), was expressed in E. coli.
- Molecular Mass
- The 68 amino acid recombinant protein has a predicted molecular mass of approximately 7.851 kD. The DTT-reduced protein migrates at approximately 7 kD and the non-reduced protein migrates at approximately 9 kD by SDS-PAGE. The N-terminal amino acid is Ser.

- Storage & Handling
- Upon receipt, store unopened vials between 2°C and 8°C immediately and use within 12 months from date of receipt. Prior to use, reconstitute the lyophilized powder with 0.2 ml of PBS containing a carrier protein (e.g., 1% BSA, protease free), pH 7.4. Re-cap vial, vortex. Allow the reconstituted standard to sit at room temperature for 15 minutes, vortex again to mix completely. The reconstituted standard stock solution can be aliquoted into polypropylene vials and stored at -70°C for up to one month. Do not re-use diluted standards. Use a manual defrost freezer and avoid repeated freeze thaw cycles.
ELISA - Quality tested
- Recommended Usage
Each lot of this protein is quality control tested by ELISA assay. For use as an ELISA standard, a standard curve comprised of two-fold dilutions from 500 pg/mL to 7.8 pg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
- Application Notes
This CCL5 protein is useful as a standard for a human CCL5 sandwich ELISA, using unlabeled J047C5 antibody (Cat. No. 526402) for capture and biotinylated Poly5197 antibody (Cat. No. 519703) for detection.
Note: For testing human CCL5 in serum, plasma or cell culture supernatant, LEGEND MAX™ Human CCL5 (RANTES) ELISA Kit with Pre-coated Plates (Cat. No. 440807 & 440808) are specially developed and recommended.
RANTES/CCL5 is secreted by lymphocytes, macrophages, eosinophils, platelets, and fibroblasts. RANTES is expressed by hepatocytes, sinusoidal endothelial cells and biliary epithelium during chronic hepatitis C.
- RANTES can promote IL-2 and IL-5 expression in human T cells, and the expression of IL-6 and IL-8 in fibroblasts from patients with RA. RANTES modulates the LPS-induced IL-6 and TNFα in peripheral blood monocytes; this modulation is through the induction of IL-10 by RANTES.
- RANTES/CCL5 is chemoattractant for memory and activated T cells, monocytes, dendritic cells, eosinophils, NK cells, mast cells, and basophils.
- CCL5 binds to CCR1, CCR3 and CCR5 receptors. In addition CCL5 binds to CCR9; nevertheless, it does not induce calcium flux and therefore no chemotactic response.
- Bioactivity was measured by its property to chemoattract T cells in a dose dependent manner. The ED50 is 5-15 ng/ml.
