Total protein lysates (15 µg protein) from human serum (lane 1), mouse serum (lane 2), human IgG1 recombinant antibody (lane 3) and human IgG2 recombinant antibody (lane 4) were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to nitrocellulose, and probed with 1 µg/mL (1:500 dilution) of purified anti-human IgG1 Fc antibody, clone M1308A10 (upper panel). Proteins were visualized by chemiluminescence detection (Cat. No. 426303) using a 1:3000 diluted anti-rat-IgG secondary antibody conjugated to HRP for purified anti-human IgG Fc antibody. Ponceau S staining (lower panel) was used as loading control for human and mouse serum. Lane M: Molecular weight ladder.

Description

IgG Fc is a homodimer that is composed of the constant region of the two heavy chains that form the IgG molecule. The Fc fragment mediates opsonization, antibody dependent cellular cytotoxicity (ADCC) and complement activation through the binding to Fc receptors such as CD16, CD32, CD64 and the complement factor C1. Clone M1308A10 was generated by immunogen derived from recombinant human IgG1 Fc fusion protein and only recognizes human IgG1 Fc heavy chain, but not human IgG2 Fc heavy chain.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone M1308A10 was generated by immunogen derived from recombinant human IgG1 Fc fusion protein and only recognizes human IgG1 Fc heavy chain, but not human IgG2 Fc heavy chain.
