Western blot of HRP anti-SNAP-25 antibody (clone SP12). Lane 1: Molecular weight marker; Lane 2: 20 µg of human brain membrane lysate; Lane 3: 20 µg of mouse brain membrane lysate; Lane 4: 20 µg of rat brain membrane lysate. The blot was incubated with 0.005 µg/mL of the primary antibody overnight at 4°C. Enhanced chemiluminescence was used as the detection system. -
IHC staining of HRP anti-SNAP-25 antibody (clone SP12) on formalin-fixed paraffin-embedded human brain tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with 2 µg/mL of the primary antibody overnight at 4°C. DAB was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm -

Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with 0.5 µg/mL of the primary antibody overnight at 4°C. DAB was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm -

SNAP-25, also known as synaptosomal-associated protein 25, is a component of the trans-SNARE complex. It is a Q-SNARE protein anchored to the membrane on the cytosolic face, and does not contain a trans-membrane domain. The SNARE complex is involved in the vesicle fusion process. SNAP-25 is implicated in the inhibition of voltage-gated calcium channels, and has been shown to decrease calcium responsiveness in glutamatergic synapses.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.001 - 0.02 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 0.5 - 2.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
