TE-71 mouse thymic epithelial stromal cell line cells were fixed with 1% paraformaldehyde (PFA) for 10 minutes and blocked with 5% FBS for 30 minutes. Then the cells were stained with 5 µg/ml podoplanin (clone 8.1.1) Alexa Fluor® 594 (red) in blocking buffer at room temperature for 3 hours. Nuclei were counterstained with DAPI (blue). The image was captured with a 20X objective.

    C57BL/6 mouse frozen kidney section was fixed with 4% paraformaldehyde (PFA) for 10 minutes at room temperature and blocked with 5% FBS for 30 minutes at room temperature. Then the section was stained with 10 µg/ml CD100 (clone BMA-12) purified and podoplanin (clone 8.1.1) Alexa Fluor® 594 (red) overnight at 4°C, followed by 2.5 µg/ml of goat anti rat IgG (clone Poly4054) Alexa Fluor® 647 (green) for 2 hours at room temperature. The image was captured by 10X objective.

Description

The mucin-type glycoprotein podoplanin is thought to be involved in the development of the lymphatic vascular system. Podoplanin is named after its expression in the kidney glomerular epithelial cells (podocytes). It has a potential role in tumor progression.

Recommended Usage

Each lot of this antibody is quality control tested by immunocytochemistry. For immunocytochemistry, a concentration range of 2.0 - 10 μg/ml is recommended. For immunohistochemical staining on frozen tissue sections, the suggested use of this reagent is 5.0 - 10.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 594 has an excitation maximum of 590 nm, and a maximum emission of 617 nm.
