Experimental Design

Cells used: SUM159
Brief Protocol:
Cells were trypsinized, neutralized with DMEM 10%, and re-suspended in ice-cold PBS with 2% IFS at 1x10^6 cells per 100 uL. The cells were incubated on ice with 1:100 antibody for 30 min, washed with 9 times the original volume of ice-cold PBS- with 2% IFS, and re-suspended in 500 uL ice-cold PBS- with 2% IFS.
Results Summary: 10.3% of the cells were GD2 high.

Technical Service Notes

Trypsinization can affect epitopes as it is enzyme-based. This can make it difficult for antibodies to recognize their targets.
