Excellent cell tracer dye for CD8 T cell proliferation in vitro.

Experimental Design

Application:	Proliferation Assay/FACS
Cells used:	In vitro cultured CD8 T cells with DCs.
Brief Protocol:	1. Primary splenic CD8 T cells were isolated from mouse spleen. 2. CD8 T cells were diluted in RPMI medium (without FCS!!) to a concentration of 1 ∙ 107 cells per ml. 3. 1 µL of 5 mM CFSE stock solution per ml cells suspension was used. 4. Cells were incubated at 37 °C for 20 min. 5. Staining was stopped by adding 10 times the original staining volume of RPMI medium containing 10 % FCS.6. After centrifugation cells were used for proliferation assay in vitro.
Results Summary:	Excellent cell tracer dye to follow cell divisions in cell culture in vitro.
