Weakly Stains Surface CD1d on Mouse Splenic Leukocytes

CD1d expression was only weakly visualized by this antibody.
Experimental Design

Application: Flow cytometry
Cells used: Mouse splenic leukocytes
Brief Protocol: Isolate splenocytes. Wash cells once in FACS buffer (PBS plus 1% BSA or serum). Block Fc receptors and stain for viability in PBS for 20 minutes on ice. Wash cells once in FACS buffer. Stain with isotype control or CD1d PerCP Cyanine5.5 antibody (1:500 dilution, 100 uL per million cells) for 20 minutes on ice in the dark. Wash cells once in FACS buffer. Resuspend cells in 100-250 uL FACS buffer containing 1.5% paraformaldehyde.
Results Summary: CD1d expression was difficult to distinguish from isotype control.
Additional Notes: PerCP Cyanine5.5 is a fairly unstable conjugate, particularly compared to other available fluorophore/antibody conjugates.

Technical Service Notes

This product's lot expired in March of 2014. That may indicate why you're observing weak staining. We'd suggest trying a freshly produced lot to see if this contributes to better staining.This product is also recommended to be used at a 1:80 dilution as noted on the data sheet. The product, of course, can be titrated down from there.
