Experimental Design

Application:	Flow cytometry
Cells used:	Mouse splenocytes
Brief Protocol:	Resuspend in PBS/BSA and count the cells. Load 1x106 cells onto a V-Bottom 96-well-plate. Spin down, resuspend in 50uL of PBS/BSA containing αFcγR (24.62) 30 µg/ml and incubate 15min at 4°C. Spin down and resuspend in 50 µl antibody staining solution containing anti-mouse CD21/35 (CR2/CR1) PE/Cyanine7 (1/400). Incubate 15min at 4°C in the dark. Wash twice with PBS/BSA, resuspend and transfer to FACS tubes to read. Use PI to exclude the dead cells.
Results Summary:	5-10% of live B cells are CD21/35hiCD23lo MZ B cells in a naïve spleen.
