14-3-3 Induced Intracellular Signaling


The 14-3-3 family of proteins includes seven different isoforms. Each protein serves as an adaptor molecule and can bind to transcription factors, enzymes, kinases, phosphatases, and other signaling molecules through a Ser/Thr motif. Binding of 14-3-3 can regulate the location, stability, activity, and phosphorylation status of its binding partners making it an important regulator of cellular functions. Among the hundreds of identified binding partners, 14-3-3 can bind to and regulate PI3K, AKT signaling targets including BAD and YAP, JNK, and MEK/ERK to regulate cell growth, proliferation, and apoptosis through these pathways.
