    Human paraffin-embedded breast cancer tissue slices were prepared with a standard protocol of deparaffination and rehydration. Antigen retrieval was done with Citrate-Buffered 1X pH6.0 at 95°C for 40 minutes. Tissue was washed with PBS/ 0.05% Tween20 twice for five minutes and blocked with 5% FBS and 0.2% gelatin for 30 minutes. Then, the tissue was stained with 10 µg/ml of Purified anti-human Podoplanin (Clone LpMab-21) antibody overnight at 4°C. On the next day, tissue was incubated with Alexa Fluor® 647 Goat anti-mouse IgG2a (clone RMG2a-62) antibody (red) and Alexa Fluor® anti-human DCR3 (Clone B08-35) antibody (green). Nuclei were counter-stained with DAPI (blue). The image was scanned with a 10X objective and stitched with MetaMorph® software.

Description

Podoplanin is a 40-43 kD type-I transmembrane sialomucin-type glycoprotein, also known as T1a, gp36, gp38, gp40, and Aggrus. Originally detected on the surface of podocytes, futher characterization showed podoplanin has a broad tissue distribution, including mesothelial cells, epithelial cells, follicular dendritic cells, and a variety of tumor cells. It has been reported that podoplanin is the ligand of CLEC2 and is involved in lymphatic vessel formation, platelet aggregation, and tumor metastasis. Podoplanin may serve as a useful marker for tumor diagnosis and prognosis.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µl volume. For immunohistochemistry on formalin-fixed paraffin-embedded tissue, a concentration range of 5.0 - 10 µg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
