Whole cell extracts (15 µg protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-PPP5 Antibody, clone O95G5, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular Weight marker.

    Whole cell extracts (300 µg total protein) prepared from HeLa cells were immunoprecipitated overnight with 2.5 µg of Purified Mouse IgG1, κ Isotype Control Antibody (Cat. No. 400102) or Purified anti-PPP5 Antibody, clone O95G5. The resulting IP fractions and whole cell extract input (5%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with a 1.0 µg/mL (1:500 dilution) of O95G5. Description

Protein phosphatase 5 catalytic subunit (PPP5) is a member of the phosphoprotein phosphatase family of enzymes and shows specificity towards phosphorylated serine and threonine residues. PPP5 displays broad substrate recognition and regulates diverse signal transduction cascades, including hormone and stress response pathways. PPP5 contains an autoinhibitory tetratricopeptide repeat (TPR) domain; PPP5 activity is augmented by Hsp90A, which releases TPR inhibition of the enzyme and also promotes the interaction of the PPP5 catalytic domain with client proteins.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg per ml. For immunoprecipitation, the suggested use of this reagent is 2.5 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone weakly reacts with two unidentified proteins of ~45 and 50 kD. This protein was also detected in O95G5 immunoprecipitates from HeLa extracts. We therefore do not recommend using this clone for co-immunoprecipitations.
