Description

TAB1 (TGFß-Activated Kinase 1-Binding Protein 1), also known as TAK1-Binding Protein 1, is a regulator of the MAP kinase kinase kinase MAP3K7/TAK1, which mediates MAPK signaling pathways and proinflammatory signaling induced by TGF-β, IL-1, TNF-α and WNT-1. In the signaling pathway triggered by TGF- β, the binding of TAB1 and TAK1 triggers TAK1 autophosphorylation and activation. In the response to TNF-α or IL-1, TAK1 binds with  TAB1/TAB2 or TAB1/TAB3 to form distinct complexes. In addition, TAB1 can also activate MAPK14/p38α, which represents an alternative activation pathway upon various stimuli. Alternatively spliced transcript variants which encode distinct isoforms have been reported. On the other hand, TAB1 gene disrupted mice demonstrate reduced TAK1 activity and are embryonic lethal.

Recommended Usage
