    Total cell lysates (15 µg total protein) from Jurkat cells untreated (-) and treated (+) with anisomycin (25 µg/mL for 15 minutes) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-p38 MAPK Phopsho (Thr180/Tyr182) Antibody, clone Poly29035, at a 1:500 dilution overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP donkey anti-rabbit IgG Antibody (Cat. No. 406401) at a 1:3000 dilution. Purified anti-p38 MAPK Antibody, clone Poly6224 (Cat. No. 622402), and Direct-Blot™ anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:500 dilution. Lane M: Molecular Weight marker.

Description

The p38 MAP kinase is a stress-activated protein kinase. The p38 MAP kinase is activated by treatment of cells with proinflammatory cytokines (e.g. TNF and IL-1) or by exposure of cells to environmental stress (e.g. UV radiation and osmotic shock). This activation results in the phosphorylation of residues Thr180 and Tyr182. This antibody is specific for the 39 kD p-38 protein phosphorylated at Thr180/Tyr182 It is purified by sequential chromatography on protein A and phospho- and dephospho- peptide affinity columns.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a dilution range of 1:1 is suggested. For Western blotting, the suggested use of this reagent is 1:1000 dilution. It is recommended that the reagent be titrated for optimal performance for each application.
