Whole cell lysates (15 µg) from the listed cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with a 1.0 µg/mL (1:500 dilution from 0.5 mg/mL) of purified anti-METTL14 antibody, clone W181219, overnight at 4°C. The following day, protein was chemiluminescently detected via a 1 hour room temp. incubation with HRP-conjugated Goat anti-rat (1:3000 dilution; for in-house target) and HRP-conjugated Goat anti-rabbit (1:3000 dilution) followed by imaging.

    Whole Cell Lysates (250 µg) were prepared from MOLT4 cells and then immunoprecipitated overnight with 2.5 µg of purified rat IgG2a isotype control antibody or purified METTL14 antibody (clone W181219). The resulting fractions and 5% whole cell lysate input were resolved by 4-12% Bis-Tris Gel Electrophoresis and probed with an anti-rat secondary antibody against the target.

Description

METTL14 forms a heterodimer with METTL3 called the m6A-methyltransferase complex (MTC) to methylate adenosine residues at the N6 position of eukaryotic mRNAs. MTC-induced methylation takes place at the 5'-[AG]GAC-3' consensus sites of some mRNAs. In the MTC complex, METTL14 constitutes the RNA-binding scaffold that recognizes the substrate. Once methylated, the mRNAs are recognized by distinct proteins known as “readers” that will affect their stability, splicing, and/or translation.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.025 - 1.0 µg per ml. For immunoprecipitation, the suggested use of this reagent is 2.5 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

While this clone is able to immunoprecipitate METTL14 from whole cell extracts, the resulting fraction was not significantly enriched in METTL14; therefore, this clone may not be suitable for detecting transient or low-abundance complexes.
