Total cell lysates (15 µg total protein) from Human and mouse brain tissue were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL of Purified anti-IL34 antibody, clone C051-77, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker.

    Immunoprecipitation/Western blot analysis using the anti-IL-34 antibody (C051-77). 100 ng/mL of recombinant mouse IL-34 was tested. Lane 1 was immunoprecipitated with rat IgG control antibody and lane 2 was immunoprecipitated with anti-IL-34 antibody (C051-77). Western blot analysis was performed using goat anti-mouse IL-34 antibody.

Description

IL-34 is a glycoprotein that plays important roles in the proliferation, survival, and differentiation of monocytes and macrophages. IL-34 is expressed in the heart, brain, lung, liver, kidney, thymus, spleen, and other organs. It specifically binds to CD115/M-CSF receptor (CSF-1 receptor or c-Fms) and activates ERK1/2 phosphorylation. The function of IL-34 overlaps with CSF-1 to regulate myeloid lineage development. IL-34 also promotes inflammatory processes and regulates osteoclast proliferation, differentiation, and bone reabsorption.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1.0 µg per ml. For immunoprecipitation, the suggested use of this reagent is 2 - 5 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

The theoretical MW of IL-34 is 27 kD, the observed MW is 50 kD.
