Total cell lysates (15 µg total protein) from HeLa and NIH/3T3 cells (negative controls) and mouse and human brain tissue were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of purified anti-GFAP antibody (clone 15C7D5D2) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:10000 dilution (lower). Lane M: Molecular weight marker. Predicted GFAP expression was obtained from Human Protein Atlas.

Description

GFAP is an intermediate filament protein coexpressed with vimentin in mature astrocytes. Intermediate filaments (IFs), together with microtubules and microfilaments, form an interconnected network of the cytoskeleton, which gives cells their form, shape and functions. This gene encodes one of the major intermediate filament proteins of mature astrocytes. It is used as a marker to distinguish astrocytes from other glial cells during development. Mutations in this gene cause Alexander disease, a rare disorder of astrocytes in the central nervous system. The appearance of GFAP breakdown products in serum and CSF have been linked to traumatic brain injury (TBI).

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.05 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Multiple GFAP protein fragments ranging from 38 to 48 kD have been reported in human CNS lysates resulting from caspase- and calpain-mediated cleavage of GFAP.
