Total cell lysates (15 µg total protein) from HEPG2 (negative control), A549 (negative control), SR and RPMI8226 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 2.0 µg/mL of Purified anti-Blimp1 Antibody, clone ROS195G, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker.

Proteins were visualized using a goat anti-mouse IgG secondary antibody conjugated to HRP and chemiluminescence detection. Purified anti-GAPDH antibody was used as a loading control.

Description

Blimp-1 is a 98 Kd protein containing five Kuppel-type zinc finger domains that confer sequence specific DNA binding.  It has been postulated to be a master regulator of terminal B cell differentiation. In the BCL-1 lymphoma model of differentiation from mature B cells to plasma cells, ectopic expression of Blimp-1 is sufficient to cause terminal differentiation manifested by loss of surface immunoglobulin, IgM secretion, expression of syndecan-1 and cessation of cell division. Recently, it is reported that Blimp-1 is also involved in T cell differentiation and T cell homeostasis and suppression of IL-2 production.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 5 µg per mL. It is recommended that the reagent be titrated for optimal performance for each application.
