Description

CD3 (Clone SK7): The CD3/T-cell receptor (TCR) complex is composed of two CD3ε, one CD3γ, one CD3δ, one CD3ζ (CD247), and a T-cell receptor (α/β or γ/δ) heterodimer. It is found on all mature T cells, NK T cells, and some thymocytes. It plays a role in antigen recognition, signal transduction, and T cell activation.

CD45 (Clone 2D1): It is a type I membrane glycoprotein also known as leukocyte common antigen (LCA) and T200. It functions as a tyrosine phosphatase expressed on the plasma membrane of all hematopoietic cells, except erythrocytes or platelets. CD45 regulates a variety of cellular processes including cell growth, differentiation, cell cycle, and oncogenic transformation.

CD56 (Clone 5.1H11): A single transmembrane glycoprotein, also known as NCAM (neural cell adhesion molecule), Leu-19, or NKH1. It is expressed on NK and NKT cells, but in other cells and tissues as well, such as large granular lymphocyte (LGL) leukemias, small-cell lung carcinomas, neuronal derived tumors, myelomas, myeloid leukemias, brain cerebellum and cortex, and neuromuscular junctions. CD56 plays a role in homophilic and heterophilic adhesion via binding to itself or heparan sulfate.

CD16 (Clone B73.1): Also known as low affinity IgG receptor III (FcγRIII), it is expressed as two distinct forms (CD16a and CD16b). CD16a (FcγRIIIA) is expressed on the surface of NK cells, activated monocytes, macrophages, a subset of T cells, and placental trophoblasts in humans. CD16b (FcγRIIIB) is expressed specifically on neutrophils. CD16 binds aggregated IgG or IgG-antigen complex which functions in NK cell activation, phagocytosis, and antibody-dependent cell-mediated cytotoxicity (ADCC).

CD19 (Clone SJ25C1): Also known as B4, it is a member of the immunoglobulin superfamily expressed on B cells (from pro-B to blastoid B cells, absent on plasma cells) and follicular dendritic cells. CD19 is involved in B cell development, activation, and differentiation. CD19 forms a complex with CD21 (CR2) and CD81 (TAPA-1), and functions as a BCR co-receptor.

Storage & Handling
The antibody solution should be stored undiluted between 2°C and 8°C, and protected from prolonged exposure to light. Do not freeze.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 20 µl per million cells or 20 µl per 100 µl of whole blood.

Application Notes

This cocktail is intended to differentiate and determine the frequencies of T-Cells, B-Cells, NK-Cells and NKT-Cells in a single test format using either lyse-wash or lyse-no wash human peripheral blood assays. It may be used as a stand-alone product or as a common marker backbone to which additional antibodies may be added for specific applications.

Note: Isotype controls are not included. Single color compensation controls are not included.
