Chromatin Immunoprecipitations (ChIP) were performed with cross-linked chromatin samples from 4 X 106 of Jurkat cells with either A)1:100 dilution of Go-ChIP-Grade™ Purified anti-HDAC1 (Clone 10E2/HDAC1, Cat. No. 815103) or B) equal amount of Purified Mouse IgG1, κ Isotype Control Antibody (Clone MG1-45, Cat. No. 401401) by using Go-ChIP-Grade™ Protein G Enzymatic Kit (Cat. No. 699904). The enriched DNA was purified and quantified by real-time qPCR using primers targeting human CDKN1A gene region or α-Satellite repeats. The amount of immunoprecipitated DNA in each sample is represented as signal relative to total amount of input chromatin. -

Total cell lysates (15 µg protein) from HeLa (lanes marked as 1) and NIH3T3 cells (lanes marked as 2) were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to a nitrocellulose membrane, and probed with serial dilutions of purified anti-HDAC1 antibody (clone 10E2/HDAC1) as indicated (1:500, 1 µg/mL; 1:1000, 0.5 µg/mL; 1:2000, 0.25 µg/mL; 1:4000, 0.125 µg/mL). Proteins were visualized using chemiluminescence detection after incubation with a 1:3000 dilution of HRP conjugated goat anti-mouse-IgG secondary antibody for the anti-HDAC1 (upper panel) or a 1:5000 dilution of Direct-Blot™ HRP anti-β-Actin (clone 2F1-1, lower panel). Lane M: Molecular weight ladder. Non-specific bands can be detected at high antibody concentrations.

HDAC1 is a member of class I histone deacetylase (HDACs) family, which modulates the chromatin structure by removing acetyl groups (deacetylation) from the side chain of lysine residues on the N-terminal region of core histones (H2A, H2B, H3 and H4). Generally, histone deacetylation gives a tag for epigenetic repression and plays an important role in transcriptional regulation, cell cycle programming and developmental process.
HDAC1 is a 65kD nuclear protein, operating in concert with histone acetyltransferases to control core histone acetylation in highly conserved lysine residues. HDAC activity has been associated with transcriptional repression and nucleosomal condensation. HDAC1 deacetylates RELA at residue K310, which results in the inhibition of NF-κB activity. HDAC1-deficient mice were shown to be embryonic lethal, suggesting that HDAC1 plays a crucial role during embryonic development. HDAC1 also interacts with retinoblastoma tumor-suppressor protein and this complex is a key element in the control of cell proliferation and differentiation. Together with metastasis-associated protein-2 MTA2, it deacetylates p53 and modulates its effect on cell growth and apoptosis.

Each lot of this antibody is quality control tested by ChIP Assay. The suggested dilution for ChIP application is 1:100-1:350 by volume. For immunohistochemical staining on formalin-fixed paraffin-embedded tissue sections, the suggested dilution is 1:50 - 1:100. For Western blotting, the suggested use of this reagent is 1:1000-1:4000. It is recommended that the reagent be titrated for optimal performance for each application.
