Chromatin Immunoprecipitations (ChIP) were performed with cross-linked chromatin samples from 4 X 106 of Jurkat cells with either A)1:50 dilution of Go-ChIP-Grade™ Purified anti-Bcl11b (Clone 25B6, Cat. No 650603) or B) equal amount of Purified Rat IgG2a, κ Isotype Control Antibody (Clone RTK2758, Cat. No 400501) by using Go-ChIP-Grade™ Protein G Enzymatic Kit (Cat. No 699904). The enriched DNA was purified and quantified by real-time qPCR using primers targeting human CDKN1A gene region or &alpha-Satellite repeats. The amount of immunoprecipitated DNA in each sample is represented as signal relative to total amount of input chromatin. -

Total lysates (15 µg protein) from Jurkat (lane 1), HeLa (lane 2, negative control), and EL4 cells (lane 3) were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with 1:1000 diluted (0.5 µg/mL) Purified anti-Bcl11b Antibody, clone 25B6 (upper). Proteins were visualized by chemiluminescence detection using a 1:3000 diluted goat anti-rat-IgG secondary antibody conjugated to HRP for the anti-Bcl11b Antibody or 1:5000 diluted Direct-Blot HRP anti-β-Actin Antibody, clone 2F1-1(lower). Lane M: Molecular weight ladder. -
Jurkat cells were stained with anti-bcl11b, clone 25B6 (1:200 dilution), and tubulin-α, clone TU-01, followed by DyLight™ 549 conjugated anti-rat IgG and DyLight™ 488 conjugated anti-mouse IgG. Nuclei are stained with DAPI.

BCL11B is a C2H2-type zinc finger protein and is closely related to BCL11A whose translocation may be associated with B-cell malignancies. BCL11B is expressed in thymus, mainly T cells. It is indispensable for T lineage development. When BCL11B was deleted, T cells from all developmental stages acquired NK cell properties and concomitantly lost or decreased T cell-associated gene expression. These induced T-to-natural killer (ITNK) cells, which were morphologically and genetically similar to conventional NK cells, killed tumor cells in vitro, and effectively prevented tumor metastasis in vivo. Therefore, ITNKs may represent a new cell source for cell-based therapies. BCL11B is also known to be a transcriptional repressor, and is regulated by the NURD nucleosome remodeling and histone deacetylase complex. Recent research suggests that BCL11B is crucial for ameloblasts (the cells that produce tooth enamel) to form and work properly.

Each lot of this antibody is quality control tested by ChIP Assay. The suggested dilution for ChIP application is 1:50-1:100 by volume. For western blotting, suggested working dilution(s): Use 0.5 µg antibody per ml antibody dilution buffer for each mini-gel. For immunofluorescence microscopy: Use a dilution of 1:200. The optimal concentration should be determined by titration for each individual assay of interest.
