NC-08 used for IHC stainings on PFA-fixed paraffin-embedded human glioblastoma cells transplanted into murine brains.

Experimental Design

Application:	Immunohistochemistry
Cells used:	Human glioblastoma cells
Brief Protocol:	Deparaffinize and hydrate Xylene 2 x 10 minutes 100% ethanol 2 minutes 95% ethanol 2 minutes 90% ethanol 2 minutes 80% ethanol 2 minutes 70 % ethanol 2 minutes 50 % ethanol 2 minutes Antigen retrieval Boil in citrate buffer or Boil in EDTA for 15min or Incubate with Proteinase K Block with 3% H2O2 for 10min Block with blocking buffer 1h RT (10% goat serum, 1%BSA in PBS) Incubate 1st antibody 1:50 (and 1:250 not shown) o/n 4°C Incubate 2nd antibody (biotinylated) 1:200 1h RT Use Vectastatin kit to amplify staining Perform stainig with DAB Counterstain with hematoxylin Dehydrate and mount with eukitt
Results Summary:	Staining does not work properly.
Additional Notes:	As a control staining I used the D2-40 antibody which works well and shows the expected result.
