Experimental Design

Application:	Flow cytometry
Cells used:	Mouse peritoneal exudate cells
Brief Protocol:	Obtain mouse peritoneal cells by injecting and recovering PBS from a euthanized mouse peritoneal cavity. Purify CD11b+ cells using magnetic anti-CD11b bead positive selection. Stain in 100ul FACS buffer (1x PBS + 2% FBS + 2mM EDTA) + 1ul biotin anti-mouse CD11b on ice for 20 minutes. Wash 2x FACS buffer. Fix cells overnight in 2% paraformaldehyde in PBS pH 7.4. Wash 2x FACS buffer. Stain in 100ul FACS buffer + 1ul APC-Cyanine7-streptavidin. Wash 2x FACS buffer. Resuspend in FACS buffer for FACS analysis.
Results Summary:	Biotinylated antibody is clearly labelled.
