Description

CCR6 is the only defined receptor for CCL20. Therefore, CCL20 expressed by epithelial cells during infection recruits immature CCR6+ dendritic cells (DC). After, these cells down-regulate CCR6 expression and up-regulate CCR7 as they migrate to nearby lymph nodes. CCL20 competes for receptor binding to CCR6 with β-defensins -1 and -2, and other cationic antimicrobial peptides in mice and humans. Interestingly, CCL20 itself possesses potent antimicrobial activity.
CCL20/CCR6 interactions play an important role in the pathogenesis of encephalomyelitis and psoriasis in animal models of chronic inflammatory skin disorders. CCL20 is abundantly found in psoriatic plaques characterized by epidermal hyperproliferation and immunocyte infiltration. The production of CCL20 by keratinocytes could be an initial trigger for Th17 activation and a key component in maintaining the self-perpetuating cycle of Th17 activation in psoriasis.

Molecular Mass
The 70 amino acid recombinant protein has a predicted molecular mass of approximately 8.0 kD. The DTT-reduced protein migrates at approximately 11 kD and non-reduced protein at approximately 13 kD by SDS-PAGE. The N-terminal amino acid is Ala.
Purity
>98%, as determined by Coomassie stained SDS-PAGE.
Formulation
Lyophilized in sterile-filtered PBS, pH 7.2, containing 1% BSA, 0.09% sodium azide, and protease inhibitors.

Storage & Handling
Upon receipt, store unopened vials between 2°C and 8°C immediately and use within 12 months from date of receipt. Prior to use, reconstitute the lyophilized powder with 0.2 ml of PBS containing a carrier protein (e.g., 1% BSA, protease free), pH 7.4. Re-cap vial, vortex. Allow the reconstituted standard to sit at room temperature for 15 minutes, vortex again to mix completely. The reconstituted standard stock solution can be aliquoted into polypropylene vials and stored at -70°C for up to one month. Do not re-use diluted standards. Use a manual defrost freezer. Avoid repeated freeze thaw cycles.

ELISA - Quality tested

Recommended Usage

Each lot of this protein is quality control tested by ELISA. For use as an ELISA standard, a standard curve comprised of doubling dilutions from 500 pg/mL to 7.8 pg/mL is suggested. It is recommended that the reagent be titrated for optimal performance.

Application Notes

This CCL20 (MIP-3α) protein is useful as a standard for human CCL20 (MIP-3α) sandwich ELISA, using unlabeled L2412F11 antibody as capture and biotinylated L2418G3 antibody as detection.

CCL20 is expressed in activated cells, including monocytes, T cells, endothelial cells, fibroblasts, keratinocytes, synoviocytes, dendritic cells, pulmonary and intestinal epithelial cells, and dermal microvascular endothelial cells.

Function
CCL20 chemoattracts immature DC to sites of injury. CCL20 is upregulated by mediators of inflammation such as TNF or LPS. Also, CCL20 is induced by Th17 cytokines (IL-17A, IL-22, and TNFα) in keratinocytes. CCL20 expression is inhibited by IFNγ and IL-4 in synoviocytes.

T cells, Th17, Tregs, B cells, DC subsets including CD11b+CD8α-myeloid DCs, Langerhans cells (LCs), CD34+ cell-derived immature myeloid DCs, and immature monocyte-derived DCs.
