Total cell lysate from NTERA-2 cells (lane 1, 15 µg), NF-1 cells (lane 2, 15 µg) and HeLa (lane 3, 15 µg) were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with purified anti-Nanog antibody (clone 23D2-3C6). Proteins were visualized using an HRP Goat anti-mouse IgG Antibody and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin antibody (clone 2F1-1) was used as a loading control.

    NTERA-2 cells were fixed with 4% paraformaldehyde (PFA) for 10 minutes, permeabilized with 0.5% Triton X-100 for 5 minutes, and then blocked with 5% FBS for 30 minutes. Description

Nanog is a transcription factor that plays key roles in the maintenance of pluripotency and self-renewal in embryonic stem cells. Nanog expression is regulated by other transcription factors, which include Sox2 and Oct-4. It also plays a role in epithelial malignancy and is enriched in cancer stem cells. The ablation of nanog is sufficient to reduce the cancer stem cell pool.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. For flow cytometric staining, the suggested use of this reagent is ≤ 0.175 µg per million cells in 100 µl volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone is not recommended for ChIP (Chromatin Immunoprecipitation) assays (as determined by in-house testing).
