Lane 1: Molecular weight marker; Lane 2: 20 µg of human brain membrane lysate; Lane 3: 5 µg of mouse brain membrane lysate; Lane 4: 5 µg of rat brain membrane lysate. The blot was incubated with 1 µg/ml of the primary antibody overnight at 4°C, followed by incubation with HRP Streptavidin (Cat. No. 405210). Following antigen retrieval using Sodium Citrate H.I.E.R, the tissue was incubated with 5 µg/ml of the primary antibody overnight at 4°C. For detection, the HRP labeling reagent and DAB from BioLegend Ultra Streptavidin (USA) HRP Detection Kit was used (Multi-Species, component #5, DAB; Cat. No. 929901). The slide was counterstained with hematoxylin, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm

Description

Synaptotagmin 12 (Syt12) is regulated by protein kinase A (PKA) and is unique from other synaptotagmins in that it does not bind to Ca2+. Syt12 interaction with Syt1 prevents SNARE complex binding. Expression of Syt12 induces spontaneous neurotransmitter release, providing a mechanism for spontaneous synaptic-vesicle exocytosis that is Ca2+-independent.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1.0 - 10 µg per ml. For immunohistochemistry on formalin-fixed paraffin-embedded tissue, a concentration range of 5.0 - 10 µg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody is effective in immunoblotting (WB) and immunohistochemistry (IHC-P).
