In vivo Proliferation Measured with Tag-It Violet™ Labeled OT-II T Cells

OT-II CD4+ T cells labeled with Tag-It Violet™ were i.v. injected into mice followed by immunization with OVA the next day, and proliferation was assessed by proliferation dye dilution after 3 days.

Experimental Design

Application:	Cell proliferation and tracking
Cells used:	OT-II CD4+ T cells
Brief Protocol:	OT-II CD4+ T cells were isolated from spleens of OT-II transgenic mice by magnetic bead isolation. OT-II cells were labeled with Tag-It violet (5uM) for 20min at 37C, followed by quenching with cell culture medium and incubation on ice for 10min. Cell were washed and resuspended in PBS and 10x10^6 cell were i.v. injected into mice. The next day, OVA was injected intranasally. Three days later, mediastinal lymph nodes and spleen were isolated, stained with anti-CD4, and analyzed by flow cytometry.
Results Summary:	Proliferation as assessed by Tag-It dye dilution was measured in both the mediastinal lymph node and spleen.
