Brief Protocol: | After 6-day stimulation of OT-I splenocytes with Ova peptide and IL-2: Wash splenocytes with FACS buffer. Incubate cells with Fc block (1:100 dilution). Add CD8a Brilliant Violet 421™ directly to the cells (1:100 final concentration) and incubate for 30 min on ice. Wash 3x with FACS buffer. Complete FACS analysis. |
