IHC staining of purified anti-TTF-1 antibody (clone 8G7G3/1) on formalin-fixed paraffin-embedded human lung tumor tissue. Following antigen retrieval using Sodium Citrate H.I.E.R., the tissue was incubated with the primary antibody for 60 minutes at room temperature. BioLegend´s Ultra-Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. Transcription termination factor-1 (TTF-1) plays a critical role in ribosomal gene transcription. This transcription factor mediates the termination of RNA polymerase I transcription by binding to Sal box terminator elements downstream of pre-rRNA coding regions. It binds to and activates the promoter of thyroid specific genes such as thyroglobulin, thyrotropin receptor, and thyroperoxidase. TTF-1 is important in the maintenance of the thyroid differentiation phenotype.
