Recombinant human S100A8 protein (Cat. No. 719902, lane 1), S100A9 protein( lane 2) and total lysates (15 µg protein) from HeLa (negative control, lane 3) , PBMC (lane 4) were resolved by 4-20% Tris-Glycine electrophoresis, transferred to nitrocellulose, and probed with 1:5000 (0.1 µg/mL) diluted purified anti-MRP-14 (S100A9) (clone A10105J). Proteins were visualized by chemiluminescence detection using 1:3000 diluted HRP anti-mouse-IgG secondary antibody (Cat. No. 405306). 1:2000 dilution of Direct-Blot™ HRP anti-β-actin antibody (clone 2F1-1, Cat. No. 643807) was used as a loading control (lower). Lane M: Molecular weight ladder.

Description

MRP-14, also known as S100A9, belongs to the S100 family of proteins containing 2-EF-hand calcium-binding motifs. It forms non-covalently associated complexes with another S100 family member, MRP-8. MRP14 is expressed in neutrophils and monocytes; it also has been found in ileal tissue eosinophils. The S100A8/A9 complex has been shown to play a role during calcium-dependent activation as well as have antibacterial, antifungal, immunomodulating, and anti-proliferative effects. It also functions as a chemoattractant factor for neutrophils and has been considered an endogenous activator of Toll-like receptor 4. S100A9 has been identified as a marker for CD14(+) HLA-DR(-/low) MDSCs.

Application

WB - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.05 - 0.5 µg per mL (1:1000-1:10000). It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone does not recognize S100A8. The 114 amino acid recombinant S100A9 protein has a predicted Mw of ~13 kD for monomer and ~26 kD for dimer.
