IHC staining of purified anti-KCNA5 antibody (clone 2D10.D6) on formalin-fixed paraffin-embedded rat brain tissue. Following antigen retrieval using Sodium Citrate H.I.E.R. (Cat. No. 928502), the tissue was incubated with 10 µg/ml of the primary antibody overnight at 4°C. BioLegend´s Ultra-Streptavidin (USA) HRP kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 µm

The blot was incubated with 10 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system.

Description

KCNA5 is a potassium voltage-gated ion channel with 6 transmembrane spanning domains, also known as Kv1.5, belonging to the shaker-related subfamily of potassium channels. It forms tetrameric potassium selective channels that open or close based on the voltage difference across the membrane. Potassium channels contribute to a diverse set of functions depending on the cell type and localization, some of which include: regulating neurotransmitter release, heart rate, insulin secretion, smooth muscle contraction, neuronal excitability, and much more.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10 µg/ml is suggested. For Western blotting, the suggested use of this reagent is 1.0 - 10 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody is effective in immunoblotting (WB) and immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P).

Clone is less reactive in human samples in IHC-P applications.
