    Chromatin Immunoprecipitation (ChIP) was performed using Go-ChIP-Grade™ Protein G Enzymatic Kit by loading 3 µg of cross-linked chromatin samples from U266 cells treated with IFNα2 with either A) 1:200 dilution of Go-ChIP-Grade™ Purified anti-IRF4 (Clone IRF4.3E4), or B) equal amount of Purified Rat IgG1, κ Isotype Control Antibody (Cat. No. 400401). The enriched DNA was purified and quantified by real-time qPCR using primers targeting human SUB1 gene region. The amount of immunoprecipitated DNA in each sample is represented as signal relative to the 5% of total amount of input chromatin.

Description

The IRF family consists of at least nine members. IRF4 and IRF8 are highly homologous to each other and also redundant in function. IRF4 is critical for Th2 and Th17 development. Together with TRF8, it plays an essential role in macrophage and dendritic cell development and function. IRF4 is also reported to be essential for pre-B cell development, receptor editing, germinal center reactor and plasma cell differentiation.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 10 µg/ml. For ChIP applications, the suggested dilution is is 1:50-1:200 by volume. It is recommended that the reagent be titrated for optimal performance for each application.
