Western blot of purified anti-HSF2 antibody (clone 3E2). Lane 1: Molecular weight marker; Lane 2: 30 µg of HeLa lysate. The blot was incubated with 1 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system.

Heat shock factor protein 2 (HSF2) is a transcription factor that enters the nucleus and binds to heat shock factor promoter elements, consisting of a consensus tandem array sequence of three oppositely oriented "AGAAN" motifs, during heat shock in eukaryotes. Under normal conditions, it exists as a homodimer in the cytoplasm of cells where it binds to heat shock proteins and inactivates them. During high temperature cellular stress, misfolding of proteins occurs, which causes heat shock proteins to dissociate from the heat shock factor proteins and bind to the misfolded proteins. This leads to free heat shock proteins, which can then enter the nucleus and activate transcription.
