Total lysates of 15, 7, 3 µg protein (lanes 1, 2, 3 respectively) from Raw264.7 (Mouse) cells were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1:500 (1 µg/ml) purified anti-Bid antibody, clone 14F2. Proteins were visualized using chemiluminescence detection by incubation with HRP Goat anti-Rat secondary antibody (Cat. No. 405405, 1:3000 dilution). Direct-Blot™ HRP anti-β-actin antibody was used as a loading control (Cat. No. 643807, 1:8000 dilution).

BID, a BH3 interacting domain death agonist, is a member of the Bcl-2 family. Upon activation, in response to Fas/TNF-R1 death receptor signal, it is cleaved by caspase 8 and translocated to mitochondria. There it induces cytochrome C release, which in turn activates downstream caspases.
