Experimental Design

Application:	Flow cytometry
Cells used:	5 day old mouse splenocytes
Brief Protocol:	Mash spleen through nylon mesh Purify mouse B cells from spleen using magnetic separation column Wash cells in FACS buffer (1x PBS + 2% FBS + 2mM EDTA) Resuspend cells in 100 ul FACS buffer + 1 ul Alexa Fluor 700 anti-mouse CD45R/B220, + 1 ul APC anti-mouse CD93 on ice for 10 minutes Wash cells in FACS buffer and analyze on flow cytometer
Results Summary:	Neonatal spleens contain CD93+ immature B cells
