Quantifying Neutrophils in Bone Marrow (APC anti-mouse/human CD11b)

Number of neutrophils in femur were measured in C57BL/6 mice given a 6mm skin wound.
Experimental Design

Application: Flow Cytometry
Cells used: C57BL/6 Bone Marrow
Brief Protocol: Isolate bone marrow cells to a concentration of 10E7 per ml in Cell Staining Buffer (Biolegend).
Stain 100ul of cell suspension.
Add leaf anti-CD16/32 (Biolegend) to 1 ug/ml for 10 minutes.
Add 1ul of APC anti-mouse CD11b (and 1ul PE/Cyanine7 anti-mouse Ly-6G) to cell suspension.
Stain for 20' at RT in darkness.
Wash with 1ml cell staining buffer, spin at 400xG for 5 minutes.
Resuspend in 500 ul cell staining buffer.
Assess on Attune NxT (Record 200 ul at 200 ul/min)
Results Summary: After wounding, the number of neutrophils in bone marrow drops and then steadily rises over 5 days.
Additional Notes: Great separation. This antibody is good for detecting neutrophils. I have also used this antibody successfully in expanding human CD34 cells as a marker for myelopoiesis.
